Development of Pharmaceutical Abuse Deterrent Capsule Formulations
Summary: Recent regulations and public pressure are mandating more stringent abuse prevention mechanisms to be incorporated in pain management. One popular mode of abuse of oral formulations with abuse – susceptible drugs is the extraction of API from the dosage form and subsequent injection. The objective of this study was therefore to develop liquid capsule fill formulations, which are mitigated to prevent abuse by extraction and injection of the abuse – susceptible drugs. Different lipid based formulation sytems containing the active ingredient, solvent, surfactant, plasticizer, water and a gelling agent or an ion exchange resin were prepared and tested regarding extractability, syringeability, API release profile and stability in soft gelatin capsule formulations.
